Special studies have established that the disaccharide lactulose (fructose-galactoside) is an active bifidogenic factor in human milk. To optimize the formulation of milk-based baby food products, lactulose must be added to cow's milk. In addition, lactulose is widely used in many countries around the world as a preventive and therapeutic agent, especially in the case of dysbiotic phenomena. Lactulose can be used in food products as a highly soluble sweetener, in alcoholic and non-alcoholic drinks to minimize their toxic effects, as well as feed additives and in milk replacer formulations to prevent dysbiosis in young farm animals.

Lactulose can be obtained from lactose by rearranging α-glucose into fructose (Fig. 50). The mechanism of rearrangement is interpreted in different ways. In a slightly alkaline environment, glucose can be transformed into fructose by the LA-transformation mechanism through the enol form, as well as by Amadori rearrangement with the formation of the intermediate product lactosylamine. Isomerization of lactose into lactulose is carried out in an alkaline environment at pH 11... 12 by introducing 0.27 ± 0.02% sodium hydroxide in the form of a 40% solution for food syrups or 0.40 ± 0.05%

calcium hydroxide in the form of freshly prepared lime milk for feed additives. Optimal isomerization mode: temperature 85...95 °C, holding time 5...7 minutes with rapid (sharp) cooling to 15...25°C, which eliminates autocatalytic decomposition of carbohydrates and an increase in the color of the solution. If rapid cooling is not possible, the solution must be acidified to pH 7.0...8.0 with citric or lactic acid for food syrups, acid whey for feed additives. During the isomerization process, up to 30% of lactose is transformed into lactulose. The result is a lactolactulose solution containing about 7% lactulose and 15% unreacted lactose, as well as ballast substances - mineral salts (0.7%), nitrogenous compounds (0.3% nitrogen) and coloring compounds, mainly melanoidins.

Demineralization is carried out for food syrups at a temperature of 18...22 °C to a desalting level of 70...75% in electrodialysis units according to the production modes of demineralized whey. Deeper desalting, for example for baby food at a level of 90%, can be carried out by ion exchange treatment on KU-2-8 and EDE-10P resins, as in the production of milk sugar.

Refining is carried out at 20...30 °C by adding active carbon (1.5%), ground diatomite (0.15%) and sodium hydrosulfite (0.01%). The dose of reagents is calculated in relation to lactose. The solution is kept with stirring

10... 15 minutes and filtered through a “belting” cloth with a washed layer of ground diatomite. The lactolactulose filtrate is concentrated in a vacuum apparatus at a temperature of 55...65 °C to a mass fraction of dry substances of 55...60%, which corresponds to a density of 1245 ± 5 kg/m3. The condensed syrup is heated to 70 °C and sent for crystallization.

Lactose crystallization is carried out to increase the proportion of lactulose in the finished product according to the following regime: cooling with constant slow stirring of the mass at a speed of 2...3 °C per 1 hour to a temperature of 8...10 °C and then holding at this temperature for 10...12 h.

The released lactose crystals are separated by centrifugation in filter-type devices and washed with a small amount of cold water. The crystalline precipitate of milk sugar is used for dissolution in a closed cycle for the production of lactulose syrups.

If necessary (depending on consumer requirements for the quality of lactulose syrups), repeated demineralization (for baby food and pharmaceuticals), thickening (food products and drinks) with crystallization and separation of crystals is carried out. Repeated thickening is carried out to a mass fraction of dry substances of 65...70%, which corresponds to a density of 1280 ± 10 kg/m3. To intensify the crystallization process, a seed of fine-crystalline lactose is added in an amount of 0.3...0.5% of the syrup weight. The final cooling temperature is 5 °C, followed by holding for up to 10 hours, which prevents precipitation during storage of lactulose syrups.

Heat treatment of lactulose syrups is carried out at a temperature of 70...75 °C for 10...15 minutes to guarantee microbiological purity, safety and long-term storage.
